氟伐他汀对放射性肺损伤TGF-β1的影响

摘    要:目的探讨氟伐他汀(fluvastatin)对放射性肺损伤TGF-β1表达的影响. 方法将SD大鼠随机分为对照组、单纯照射组和氟伐他汀防治组,防治组于照射前1 wk以氟伐他汀20 mg*kg-1灌胃,1次*d-1, 直至实验结束.其他组灌服生理盐水.直线加速器全胸部照射,单次剂量为20 Gy,照后5,15,30和60 d剪开胸腔, 取肺组织做病理切片,并作TGF-β1免疫组化染色, 光镜下观察染色结果. 结果放疗组显示随着照射后时间延长, TGF-β1在肺组织表达逐渐增强, 氟伐他汀防治组显示TGF-β1的表达不随病情发展而增强. 与各时间点放射组相比明显减弱. 结论氟伐他汀能明显减弱TGF-β1表达,为防治放射性肺损伤提供了实验依据.

Abstract:AIM To investigate the effects of fluvastatin on transforming growth factor beta 1 (TGF beta 1) in rats with radiation induced lung injury. METHODS SD rats were divided randomly into 3 groups: normal control, irradiated control, rats irradiated and treated with fluvastatin. Two groups of rats were irradiated with x rays at a dose of 20 Gy thoracically. Seven days before the irradiation the rats in the fluvastatin group started the treatment with fluvastatin at a dose of 20 mg per day by garaging and continued to the end of the experiment. Tissues of lung were fecthed at different times. TGF beta 1 expression was measured by immunohistochemicall staining. RESULTS There was a significantly increase in the expression of TGF beta 1 in association with the increase of time in the irradiation group. The expression of TGF beta 1 in the irradiation treatment group was dim, compared with the irradiation group. CONCLUSION Fluvastatin down regulated TGF beta 1 expression, which indicated that the degree of irradiation induced lung injury was reduced.
