以藻酸钙为载体的可注射性组织工程软骨和骨研究

摘    要:目的在裸鼠背部皮下注射软骨细胞/藻酸钙复合物、骨髓基质成骨细胞/藻酸钙复合物,观察体内软骨、骨形成的可行性。方法分别从兔耳软骨和髂骨获取软骨细胞和骨髓基质干细胞,体外扩增培养,将获得的软骨细胞和骨髓基质成骨细胞与藻酸盐复合,注射于裸鼠背部皮下,以单纯藻酸钙植入作为对照组,6,12周后进行组织学检查和X线检查,观察软骨和骨的形成。结果注射软骨细胞复合物组,裸鼠背部皮下有软骨样组织形成,软骨细胞位于类似于正常软骨组织的陷窝中,注射骨髓基质成骨细胞复合物组,裸鼠背部皮下有骨样组织形成,具有骨髓腔样结构,而对照组无软骨或骨形成。结论藻酸钙可用作软骨和骨组织工程的支架材料,以藻酸钙为载体的可注射性组织工程软骨和骨是可行的。

Abstract:Objective To observe the neocartilage and new b one formation of the injected composites of carriers /cells in vivo using calcium alginate as a tissue enginee ring carrier.Method Chondrocytes were isolated from adu lt rabbit auricular cartilage,and b one marrow stromal cells(BMSC)were obtained from rabbit illium.They were incubated and expanded in vit ro respectively.Then cells were collected apart,mixed with sodium alginate so lution,and were injected by a needle on the dorsum of nude after adding calcium chloride.At 6and 12weeks after implantation,the cartilage and bone formation was evaluate d by means of gross,X-ray and histolo gical observation.Result12weeks after implantation,cartilag e formation were observed in the calc ium alginate-chondrocytes composites.Pecimen stained with histological methods demonstrated that chondroc ytes lied in the lacuae analogous to n ormal cartilage tissue,with a basop hilic ground-glass substance aroun d it.At the same time ,new mature bone was observed in the calcium alginate-BMSC composites.All the control specimens showed no neo-cartilage and new bone formation.Conclusion Calcium alginate can be used as a carr ier in cartilage and bone tissue engineering,and in-jectable neocartilage and new bone c an be created by implanting carriers/cells composites subcutaneously i nto athymic nude mice.
