姜黄素对体外培养的增生性瘢痕成纤维细胞的生物学作用

摘    要:目的:观察姜黄素体外抗增生性瘢痕的作用.方法:以增生性瘢痕成纤维细胞为靶细胞,分别用不同浓度的姜黄素培养液培养,MTT法测定细胞的增殖情况,透射电镜进行形态学检测,流式细胞仪技术结合PI及Annexin V双标记染色检测姜黄素的促凋亡作用,实时荧光定量聚合酶链反应检测Ⅰ,Ⅲ型前胶原及Bcl-2 mRNA的表达情况.结果:姜黄素对增生性瘢痕成纤维细胞的增殖有抑制作用;透射电镜观察发现,处理组凋亡细胞增多,凋亡细胞表现出典型的超微结构特征;流式细胞仪检测发现,随着药物浓度的增加,增生性瘢痕成纤维细胞的凋亡率也在逐渐升高;实时荧光定量聚合酶链反应检测结果显示,姜黄素处理后Ⅰ、Ⅲ型前胶原及Bcl-2 mRNA的表达水平均下调.结论:姜黄素具有一定的体外抗增生性瘢痕的作用.

Abstract:AIM:To evaluate the inhibitory effects of the curcumin against hypertrophic scar in vitro. METHODS: Hypertrophic scar fibroblasts (HSFB), as target cells, were exposed to different concentrations of curcumin respectively. The proliferation of human HSFB was evaluated by methyl thiazolyl tetrazolium (MTT) colorimetric assay.
