C60-AEDP对H22和S180癌细胞生长及周期的影响

目的探讨C60-AEDP对H22肝癌细胞和S180肉瘤细胞的影响及作用机制.方法体外培养H22肝癌细胞和S180肉瘤细胞,MTT法观察药物在不同浓度、不同条件和不同时间下,对细胞生长方式的影响,FCM法了解其作用机制.结果光照12 h,C60-AEDP对H22和S180肿瘤细胞有生长抑制效应,呈浓度效应关系;在非光照条件下,C60-AEDP对H22和S180肿瘤细胞有生长抑制效应,呈浓度-时间效应关系;通过FCM观察,在光照条件下,C60-AEDP可引起G1/S周期阻滞;在非光照条件下,C60-AEDP可引起M/G1周期细胞阻滞;并都可引起细胞凋亡增加,H22、S180肿瘤细胞凋亡率在光照条件下分别增加了32%、26%,而非光照条件下分别增加了9%、5%.结论在体外条件下,C60-AEDP在光照条件下和非光照条件下,均表现出对肿瘤细胞生长微弱的抑制效应.这种微弱的抑制效应可能是通过对细胞周期的影响而发挥作用.

Effect of C60-AEDP to liver cancer cell of H22 and sarcoma cell of S180

OBJECTIVE:To nvestigate the impact and mechanism of C_(60)-AEDP to liver cancer cell H_(22) and sarcoma cell S_(180). METHODS: H_(22) and S_(180) cells were incubated at 96-hole board to observe effects of the drug on cell growth by MTT method, and to investigate the mechanism by FCM method. RESULTS: Under the illumination condition for 12 h or not, it was demonstrated that C_(60)-AEDP inhibited the growth of H_(22) and S_(180) cells, and the inhibitory rate was concentration- and time-dependents. C_(60)-AEDP caused G_1/S cycle block under illumination condition and M/G_1 cycle block without illumination. The apoptosis rates of H_(22) and S_(180) cells were enhanced in the level of 32%, 26% under illumination condition, and 9%, 5% without illumination respectively. CONCLUSIONS: C_(60)-AEDP has the slightly inhibitory effect on tumour cells of H_(22) and S_(180). It may be worked by the influence on cell cycle.
