摘    要:目的:建立桑叶中芦丁含量的测定方法。方法:高效液相色谱法,以VARIAN ODS(250 mm×4.6 mm,5μm)为色谱柱,乙腈(A)-0.5%磷酸(B)做流动相,经梯度洗脱0.01 min,A-B(18∶82);4 min,A-B(21∶79);14 min,A-B(8∶92);17 min,A-B(18∶82);18 min,A-B(18∶82)],流速1.0 ml/min,检测波长为358 nm,柱温为40℃,紫外检测器检测。结果:芦丁浓度5μg/ml~100μg/ml范围内呈良好的线性关系,(r=0.99996,n=5),精确度(RSD=1.9%~2.1%),平均回收率试验为102%。最低检出限为0.1μg/ml(以进量浓度计)。结论:实验结果表明该方法灵敏度高,精密度和准确度好,是一种简便、快速分析的方法。

Abstract:Objective: To establish a method for the determination of rutin in mulberry leaves by HPLC.Methods: VARIAN ODS(250 mm×4.6 mm,5 μm) chromatographic column was used. The mobile phase was acetonitrile(A)-0.5% phosphoric acid(B)with gradient elution 0.01 min,A-B(18∶82);4 min,A-B(21∶79);14 min,A-B(8∶92);17 min,A-B(18∶82);18 min,A-B(18∶82)].The flow rate was 1.0 ml/min.The sample size was 20 μl,the detection wavelength was at 358 nm and the column temperature was 40℃. Then HPLC UV detector was used for detection.Results: The linearity range of rutin in mulberry leaves was 5 μg/ml~100 μg/ml(r=0.99996,n=5).Relative Standard Deviation(RSD) was 1.9%~2.1%,the average recovery was 102%.The limit of detection(LOD) was 0.1 μg/ml.Conclusion: The result showed that the method was simple and fast,high sensitive,stable and accurate,which can be used for the determination of the rutin in mulberry leaves.
