摘    要:目的:制备杆菌肽脂质体,并采用HPLC法测定杆菌肽脂质体中杆菌肽含量和包封率。方法用薄膜分散法将杆菌肽制备成杆菌肽脂质体;色谱条件:色谱柱为Diamonsil-C18色谱柱(150mm×4.6mm,5μm);流动相为pH 6.0的磷酸盐缓冲液-甲醇(60∶40);柱温为25℃;检测波长为254nm;流速为1mL/min。采用低温离心法分离杆菌肽脂质体中的游离药物。结果所制备的杆菌肽脂质体的平均包封率为87.94%(RSD 1.12%, n=5);在本色谱条件下,杆菌肽在0.061~1.83μg范围内与峰面积积分值的线性关系良好(R2=0.9997),平均回收率为99.68%~101.38%。结论本HPLC法简单,准确可靠,可用于杆菌肽脂质体中杆菌肽含量及其包封率的测定。

Abstract:Objective To prepare bacitracin liposom,and to determine bacitracin content and entrapment efficiency of the liposome using HPLC method. Methods The bacitracin liposome was prepared by film dispersion method; The chromatographic conditions were Column: Diamonsil-C18 column(150mm×4.6mm,5μm); mobile phase: pH 6.0 phosphate buffer: methanol (60∶40); column temperature: 25℃; detection wavelength: 254nm; flow rate: 1mL/min.Refrigerated centrifugation using bacitracin liposomes free drug. Results The mean encapsulation efficiency of the bacitraci liposome was 87.94%(RSD 1.12%,n=5);In the chromatographic conditions bacitracin and accessories and a good separation of the solvent peak,bacitracin at 0.061-1.83μg range had a good linear relationship (R2=0.9997),recovery was 99.68%-101.38%. Conclusion The HPLC method is simple, accurate and reliable,can be used to determine bacitracin content and entrapment efficiency of the bacitracin liposom.
