摘    要:目的:探讨附睾上皮分泌的β-防御素3(HBD-3)在人精子上的表达和功能,以及是否参与弱精子症的发生。方法:采用间接免疫荧光染色和Western blot检测HBD-3在人精子的定位和表达;采用计算机辅助精子分析(CASA)检测HBD-3是否参与精子活力和前向运动能力。结果:HBD-3在弱精子症精子的表达量较正常生育组下调。精子上HBD-3表达水平与精子活力呈正相关性。抗HBD-3抗体将干扰正常精子活力,而rHBD-3可改善弱精子症精子的活力。结论:在男性生殖系统中,精子表面HBD-3参与精子活力和前向运动能力发生。本研究为男性不育症提供新型诊断靶点和治疗策略。

Abstract:Objective To investigate the expression and function of HBD-3 secreted by epididymis in human sperm and the association with asthenospermia. Methods The localization and expression of HBD-3 were detected using indirect immunostaining and Western blot. CASA was adopted to determine the function of HBD-3 in human sperm. Results HBD-3 is significantly down-regulated in asthenospermia with both reduced sperm motility (P 〈 0.05). Interference with HBD-3 function decreases sperm motility in normal sperm, while treatment with recombinant HBD-3 significantly restores HBD-3 functions in sperm from asthenospermia patients. Conclusion The present finding reveals the function of HBD-3 on sperm motlity and forward motility. It also provides novel diagnostic targets and a feasible treatment method for related male infertility.
