摘 要: | 目的:观察mAFP基因转染的树突状细胞(DC)瘤苗诱导的靶向性细胞毒性T细胞(以下简称靶向性CTL)对肿瘤细胞的杀伤作用。方法:用构建的mAFP基因转染的DC瘤苗免疫C57BL/6小鼠,取免疫小鼠脾细胞诱导CTL,LDH非放射性细胞毒性试验检测其对不同肿瘤细胞的杀伤作用。用ELISA法检测CTL诱导过程中TNF—α和IFN-γ分泌变化规律。结果:AFP基因转染的DC瘤苗免疫小鼠后能诱导具有较高抗肿瘤活性的靶向性CTL,这种靶向性CTL对AFP分泌性肿瘤细胞具有更强的杀瘤活性,且在CTL诱导过程中TNF-α和IFN-γ分泌水平明显增高,与对照组比较差异有统计学意义(P〈O.05)。结论:AFP基因重组腺病毒修饰的DC瘤苗免疫C57BL/6J小鼠,能诱导出针对AFP抗原的靶向性CTL,并也能通过分泌TNF-α和IFN-γ实现对AFP分泌性肿瘤细胞的特异性杀伤。

Abstract: | Objective: To investigate the cytolytic effect of targeting cytotoxic T lymphocyte induced by transgenie dendritic cells (DC) tumor vaccine. Methods:C57BL/6 mice were inoculated with DC tumor vaccine which was engineered with murine alpha fetoprotein (mAFP) gene. Targeting cytotoxic T lymphoeytes (CTL) was induced from spleen cells of inoculated mice and cytolytic effect was tested by LDH nonirradiated eytotoxic test. TNF-α and IFN-γ secretion level were detected with ELISA method. Results:The targeting CTL which was induced by AFP antigen has more effective eytolytic effect against AFP-secreting tumor cells than the control group, and there was remarkably significance for TNF-α and IFN-γ secretion level of the experimental group than the control group. Conclusion: Targeting CTL could be induced by transgenic DC tumor vaccine which was engineered with mAFP gene and specifically kill the tumor cells with AFP-secreting, its cytolitic effect is also related to higher level of TNF-α and IFN-γ secretion. |
