链霉亲和素产生菌培养条件的研究

摘    要:目的;通过对分泌链霉亲和素(streptavidin)的菌株L-183最佳培养条件的研究,为提高链霉亲和素的产量及其规模生产打下了基础。方法:选用3种种子培养基和5种发酵培养基,通过检测种子菌体湿质量、间接ELISA检测发酵液中链霉亲和素的效价,筛选出菌体生长快的种子培养基及链霉亲和素产量高的发酵培养基。结果:B号种子培养基、2#发酵培养基最适合L-183的生长和链霉亲和素合成;不同菌种接种量对链霉亲和素的产量也有明显影响,在140ml发酵液中,种子接种量为0.3ml,链霉亲和素产量可达111mg/L。结论:本实验筛选的培养基价格低廉,且L-183生长快、链霉亲和素产量高,可以作为规模生产链霉亲和素的首选培养基。

Study of culture conditions for streptomyces

Abstract:ObjectiveTo explore the best culture condition for L 183 which secreted streptavidin and to establish a base for a large scale production of streptavidin. MethodsThree liquid seed culture media and 5 ferment culture media were used and the best media were chosen by examining wet mass of L 183 and production of streptavidin.ResultsSeed culture medium B and ferment culture medium 2# were suitable for the growth and streptavidin synthesizing of L 183. Streptavidin production was also affected by inoculum size of the strain. Streptavidin production can reach 111 mg/L when 0.3 ml packed cells were inoculated in 140 ml culture media 2#. ConclusionThe media chosen by this study was low cost and suitable for the growth and streptavidin yield of L 183 . They were first chosen media for streptavidin production in large scale.
