小鼠急性脑损伤后微量脑脊液谷氨酸浓度的测定

摘    要:目的建立急性脑损伤后微量脑脊液谷氨酸浓度测定方法.方法采用邻苯二甲醛柱前衍生,梯度高效液相色谱荧光检测法测定急性脑损伤小鼠脑脊液中谷氨酸(glutamate, GLu)含量.结果谷氨酸的平均加样回收率为84.26%~125.6% (n=3),日内相对标准偏差(relative standard deviation, RSD)均小于4.94%(n=3),日间相对标准偏差(relative standard deviation, RSD)均小于0.78%(n=3).急性脑损伤后6 h小鼠脑脊液谷氨酸含量相对于假手术组有明显升高(P《0.01).结论本方法准确、快速,可用于微量脑脊液谷氨酸含量的测定.

Abstract:Objective To establish a method for determining glutamate concentration in microamount cerebrospinal fluid after acute cerebral insult.Methods The concentration of glutamate in cerebrospinal fluid was measured in mice after acute cerebral insult by precolumn derivatization with O-phthaldehyde.Results The average recovery of glutamate was 84.26% to 125.6%(n=3),the relative standard deviation(RSD) in one day and three days was respectively less than 4.94%(n=3),0.78%(n=3).The concentration of glutamate in cerebrospinal fluid of mice 6 h after acute cerebral insult was significantly higher than that of sham operation mice(P<0.01).Conclusion This method is accurate and rapid.The glutamate concentation in microamount cerebrospinal fluid can be determined.
