摘    要:摘要]目的:运用Meta分析方法研究MTHFR基因C677T位点多态性与口腔癌易感性的发生风险。方法:检索VIP,CNKI和PubMed数据库中有关MTHFR基因C677T位点多态性与口腔癌易感性关联研究的文献,以OR值和95%的可信区间为效应指标,应用STATA11.0软件进行Meta分析,并对发表偏倚及敏感性分析进行检验。结果:纳入7个病例对照研究,共计1318例口腔癌患者和1817例正常对照,Meta分析结果显示,总人群中,MTHFR基因C677T多态性与口腔癌风险之间没有显着关联(TVS.C:95%CI=0.688—1.272,P=0.672,Pheterogeneity=0.001;TI"VS.CC:95%CI=0.638-1.763,P=0.821,Pheterogeneity=0.059:CTVS.CC:95%CI=0.581—1.282,P=0.465,Pheterogeneity=0.002;(CT+TY)VS.CC:95%CI=0.666-1.345,P=0.761,Pheterogeneity〈0.001;TI'VS.(CC+CT):95%CI=0.709~1.247,P=0.669,Pheterogeneity=0.126)。在针对种族和对照人群来源设计的亚组分析中,来源于医院对照的亚组分析模型显示基因突变可能会降低发生口腔癌的风险。结论:MTHFR基因C677T位点多态性突变可能降低口腔癌的发病风险。

Association between MTHFR C677T polymorphism and oral cancer risk.

Abstract:Objective: To investigate the association between MTHFR C677T polymorphism and oral cancer risk. Method: The relevant literatures were search. The data of genotype distributions and other information were extracted in the groups of patients and healthy control, Odds ratio (OR) with 95 % confidence interval (CI) and publish bias was applied with STATA 11.0 software. Result: Seven published case-control studies including 1318 patients with oral cancer and 1817 controls were selected. Overall, no significant association between MTHFR C677T polymorphism and head and neck cancer risk was found in this meta-analysis (T vs. C:95 % CI=0.688-1.272,P =0.672,Pheterogeneity=0.001:TF vs. CC: OR= 1.07,95 %CI=0.638-1.763, P =0.821, Pheterogeneity=0.059; CT vs. CC: 95 % CI=0.581 -1.282, P =0.465, Pheterogeneity= 0.002: (CT+TT) vs. CC: 95 % CI=0.666-1.345, P =0.761, Pheterogeneity〈0.001 : Tr vs. (CC+ CT) : 95 % CI=0.709-1.247, P =0.669, Pheterogeneity=0.126). In the subgroup analysis ethnicity and study design, significant associations were detected in the models of hospital design with protective tendency. Conclusion: This meta-analysis demonstrates that MTHFR C677T polymorphism may be a protective factor for developing oral cancer.
