厦门市无偿献血者HTLV-Ⅰ感染率及1株病毒株膜基因分析

摘    要:目的 了解厦门市无偿献血者HTLV Ⅰ感染的阳性率、来源及基因特点 ,为在无偿献血人群中开展检测提供基础资料。方法 对厦门市无偿献血者进行抗 HTLV血清学检测 ,初筛阳性标本经WB方法确认 ,并用PCR扩增确认的 1株HTLV Ⅰ部分env片段 ,全自动测序仪测序 ,分析该株HTLV Ⅰ与HTLV Ⅰ各亚型代表株的核苷酸、氨基酸的差异性。结果 厦门市无偿献血人群HTLV Ⅰ感染率为 0 .0 1% ,该株HTLV Ⅰ的部分核苷酸序列(5 0 31~ 6 344 )和编码氨基酸序列。结论 在厦门市无偿献血者中发现的 ,该株HTLV Ⅰ来源于我国江西省 ,其核苷酸序列与现有报道病毒株序列存在较大差异 ,提示可能与其他病毒株有不同起源 ,随着国产血清学检测试剂质量的提高 ,有必要在部分地区的无偿献血人群中开展HTLV Ⅰ抗体检测。

Abstract:Objective To investigate the incidence of HTLV Ⅰ infection and analyze its genomic characteristics in blood donors of Xiamen so as to provide basic information for future HTLV screening.Method Samples from blood donors were screened by anti HTLV serological test,positive samples were tested by Western Blot;a segment of envelope gene was amplified by PCR,and the amplified segment was sequenced by auto sequencer;the differences of nucleic acid and amino acid between tested strain and standard strains of various subtypes were analyzed.Result The incidence of HTLV I infection in blood donors of Xiamen was 0.01%,and the sequence of nucleic acid(5031~6344)and amino acid from a strain of HTLV Ⅰ was obtained.Conclusion There is HTLV Ⅰ infection in the blood donors.The HTLV Ⅰ infected donor came from Jiangxi Province.The obtained HTLV Ⅰ strain,which shows much difference when compared with the reported strains, may have different origin from the reported strains.With the improvement of HTLV Ⅰ serological detection reagent in China,it is necessary to detect HTLV Ⅰ antibody in the blood donors in some areas.
