饮食对胰岛素抵抗模型大鼠肝脏脂毒性损伤的影响

摘    要:目的:观察饮食调整对模型大鼠胰岛素抵抗(IR)状态的调整作用,及对肝脏游离脂肪酸(FFA)含量和形态学的影响。方法:将24只大鼠随机分成空白组、模型组、饮食组,每组8只。空白组用普通饲料喂养,其余两组采用高脂高糖高盐饲料喂养复制IR模型成功后,空白组继续给予普通饲料喂养;模型组继续给予高脂高糖高盐饲料饲养;饮食组则调整为普通饲料饲养。2周后处死检测空腹血糖(FBG)、空腹胰岛素(INS)、计算胰岛素敏感指数(ISI)和肝脏FFA的含量,并做肝脏病理切片,观察饮食调整对IR大鼠肝脏脂毒性损伤的改善程度。结果:①与空白组比较,模型组大鼠FBG、INS显著升高,ISI显著降低(均P<0.01),与模型组比较,饮食组大鼠FBG、INS下降(均P<0.01),ISI上升(P<0.05);②与空白组比较,模型组大鼠肝脏FFA含量显著升高(P<0.01),与模型组比较,饮食组大鼠脏FFA含量显著降低(P<0.01);③病理结果显示:模型组肝细胞存在明显的脂肪变性和水样变性,饮食组则程度较轻。结论:通过调整饮食可以改善模型大鼠的IR状态,并减轻其肝脏脂毒性损伤,其作用途径之一可能是通过减轻IR大鼠肝脏FFA的生成和堆积来实现的。

Abstract:Objective:To observe the effect of diet regulation on insulin resistance(IR)state,freefatty acid's(FFA's)concentration of Liver and liver'smorphology for experimental rats with IR.Methods:24 rats were randomly allocated into blank group,model group and dietary group.Eachgroup had 8 rats.The insulin resistance models were induced by feeding of highfat-sugar-saltforage.After models were induced successfully,blank group rats were feed by normal foragecontinually;mode group rats were fed by high—fat-sugar-salt forage continually;dietary grouprats were feed by normal forage instead.After 2 weeks,fasting blood glucose(FBG),plasmainsulin(iNs),insulin sensitivity index(ISI),FFA's concentration and morphological change ofliver were observed for each group.Results:①FBG,INS levels ofmodel group increased and ISI decreased as compared with those of blank group(P<0.01);FBG,INS levels of dietary group decreased and ISI increased as compared with those of model group(P<0.01 or P<0.05);②Theliver's FFA concentration of model group increased as compared with those of blankgroup(P<0.01);compared with model group,the FFA's concentration of liver of acupuncture group decreased significantly(P<0.01);③The results of liver's pathological sections demonstrated:there are significantly adipose and hydropic degen ration on heptic cells of model group rats,but this changment is lightly for dietary group rats.Conclusion:Diet regulation can improve the IR state of model rats and reduce the liver's fatty-toxicity—damage,one of the pathways of this effect maybe implemented by reducing the FFA's production and accumulation in liver of the experimental rats with IR.
