褪黑素受体基因1B单核苷酸多态性与妊娠糖尿病相关性研究

目的探讨褪黑素受体基因1B（MTNR1B）多态性与妊娠糖尿病（GDM）发病的关系。方法依据病例-对照设计方法,选取2011年10月～2012年5月我院GDM患者（GDM组）、正常妊娠对照（对照组）各110例作为受试对象,采用全自动生化分析仪检测受试者空腹血糖（FPG）、三酰甘油（TG）、总胆固醇（TC）等指标;应用聚合酶链反应-基因测序法对所有受试者MTNR1B基因位点rs10830963多态性进行分析。使用Logistic回归分析检测GDM的危险因素。结果两组人群的MTNR1B rs10830963 C/G等位基因频率比较差异有统计学意义（P〈0.05）。GG型等位基因的FPG水平高于CC和CG型,差异有高度统计学意义（P〈0.01）;Logistic回归分析显示TC、舒张压（DP）、FPG、口服葡糖糖耐量试验1 h后血糖（1 h PG）、基因型GG为GDM的危险因素（均P〈0.05）。结论 MTNR1B基因rs10830963多态性位点与南京地区汉族GDM具有相关性。

Correlation study between single nucleotide polymorphism of melatonin receptor 1B gene and gestational diabetes mellitus

Objective To investigate the association between single nucleotide polymorphism of melatonin receptor 1B gene（MTNR1B） and gestational diabetes mellitus（GDM）.Methods According to the case-control design method,a total of 220 subjects from October 2011 to May 2012 in our hospital were selected for the study with 110 GDM patients in GDM group and 110 normal pregnancy in control group.The automatic biochemical analyzer was used to detect the fasting blood-glucose（FPG）,triacyl glycerol（TG）,total cholesterol（TC） and other indicators;MTNR1B rs10830963 polymorphisms were analyzed by PCR and DNA sequencing.Risk factors of GDM were detected by Logistic regression analysis.Results The defference of MTNR1B SNP loci rs10830963 C/G allele frequency of two groups were statistically significant（P〈0.05）.Fasting plasma glucose levels of GG genotypes were higher than those of CC and CG type,the defferences were statistically significant（P〈0.01）;Accprdomg to Logistic regression analysis,TC,diastolic pressure（DP）,FPG,oral glucose tolerance 1 hour Test（1 h PG）,genotype GG were GDM risk factors.Conclusion MTNR1B gene loci rs10830963 polymorphism has associations with GDM in the people of the Han nationality of Nanjing region.
