己酮可可碱对新生大鼠缺血缺氧性脑病的干预作用

摘    要:目的观察己酮可可碱(PTX)对新生大鼠缺血缺氧性脑病(HIE)模型的干预作用。方法建立新生大鼠HIE模型,72只出生7d的Wistar大鼠随机分为假手术组,HIE模型组和PTX组,于缺血缺氧后72h取左脑,测定脑组织水含量、大脑皮层神经细胞内游离钙离子浓度、SOD、MDA、NOS及NO。结果与HIE模型组相比,PTX组脑组织含水量、游离钙离子浓度和MDA、NOS以及NO水平均显著降低(P〈0.05;P〈0.01;P〈0.05;P〈0.05;P〈0.05),SOD水平显著升高(P〈0.05)。结论PTX对新生大鼠缺血缺氧后脑组织有保护作用,其机制可能与PTX改善能量代谢及减少局部细胞毒性物质有关。

Abstract:Objective To observe the effects of pentoxifylline(PTX) on newborn rats with hypoxic-ischemic encephalopathy(HIE). Methods Models of 7-day-old newborn Wistar rats with HIE were established and 72 rats were randomly divided into sham operation group,HIE model group and PTX group.72h after hypoxic-ischemic operation the left side brains were taken out and the water content,intracellular calcium content(%) and the level of SOD,MDA,NOS,NO were measured. Results Compared with HIE model group,the content of water and calcium and the level of MDA,NOS,NO were markedly reduced(P<0.05,0.01,0.05,0.05,0.05),while the levels of SOD were increased significantly(P<0.05). Conclusion PTX could protect brain of neonatal rat against hypoxic-ischemic damage,which might be the result of the improvement of energy metabolism and the removal of local cytotoxic substance.
