细胞命运决定因子在乳腺癌中表达及其与cyclinDcyclinD1、c-jun和Her-2表达相关性研究

摘    要:目的探讨乳腺癌组织中细胞命运决定因子(cell fate determination factor dachshund,DACH1)表达及其与cyclin D1、c-jun和Her-2蛋白的表达相关性。方法采用免疫组织化学方法检测60例乳腺癌中DACH1、cyclin D1、c-jun和Her-2蛋白的表达,30例乳腺纤维瘤中DACH1的表达;并分析其与乳腺癌临床病理特征的关系。结果 DACH1在乳腺癌中的阳性表达率为33.3%,明显低于其在乳腺纤维腺瘤中的阳性表达率86.7%(P〈0.05)。DACH1在淋巴结阳性组中的表达低于无淋巴结转移组(P〈0.05)。乳腺癌中DACH1与Her-2蛋白的表达无明显相关性,与cyclin D1、c-jun的表达呈负相关性(r=-5.62,P〈0.05;r=-5.46,P〈0.05)。结论 DACH1表达缺失及低表达可能与乳腺癌进展有关,DACH1、cyclin D1、c-jun的异常表达预示着乳腺癌较高的侵袭性及患者预后的不良;在乳腺癌中,三者的表达具有相关性,可能和疾病的进展有一定的关系,并且可作为评估乳腺癌预后的指标。

Abstract:Objective To study the correlation between the expression of cell fate determination factor dachshund(DACH1) and the expression of cyclin D1, c-jun and Her-2 in breast carcinoma. Method Breast tissue specimens from 60 patients with breast carcinomas and 30 patients with breast fibroma were studied. Immunohistochemistry was used to detect the expressions of DACH1, cyclin D1, c-jun and Her-2, then the correlation between DACH1 and the clinicopathological features of patients with breast carcinoma was analyzed. Result The positive expression rate of DACH1 in breast fibroma tissue(86.7%) was significantly higher than that in patients with breast carcinomas(33.3%)(P〈0.05). The abnormal expression of DACH1 was associated with lymph node metastasis(P〈0.05). The expression of DACH1 protein was negatively correlated with the expression of cyclin D1 and c-jun(r=-5.62, P0.05; r=-5.46, P0.05), but it was not related with the expression of Her-2. Conclusion The absent or low expression of DACH1 may play an important role in carcinogenesis and progression of human breast carcinoma. The absent or low expression of DACH1, cyclin D1 and c-jun means higher invasiveness and poor prognosis in breast carcinoma. The expressions of the above three proteins in breast carcinoma relate to each other, indicating possible relationship with the progression of breast carcinoma.
