摘 要: | 目的研究大鼠创伤性脑水肿皮质中AQP-4与MMP-9的表达关系及甲强龙对其表达的影响。方法采用成年雄性SD大鼠随机分为甲强龙组(n=25)、模型组(n=25)、假手术组(n=5);甲强龙组和模型组均采用改进Feeney法制作脑损伤模型。甲强龙组致伤后即刻腹腔内给药,模型组则注射对应生理盐水。根据处死时间分为6h、24h、3d、5d、7d,5组。过度麻醉并断头取脑,对损伤皮层采用HE染色及免疫组化进行AQP4与MMP-9的检测。结果在模型组与甲强龙组中6h已可见AQP4与MMP-9的表达,MMP-9与AQP-4的表达均在3d达到高峰,模型组AQP4及MMP-9的表达显著高于甲强龙组(P〈0.05),相关系数(r=0.753,P〈0.01)。结论创伤性脑损伤后大鼠AQP4与MMP9的表达呈正相关;甲强龙冲击治疗能够通过下调AQP4及MMP9的表达来发挥减轻脑水肿的作用。
