酪酸梭菌联合美沙拉嗪对溃疡性结肠炎小鼠的治疗作用及其机制

摘    要:目的观察酪酸梭菌联合美沙拉嗪对小鼠溃疡性结肠炎(UC)的治疗作用,并探讨其可能的作用机制。方法用3%的葡聚糖硫酸钠(DSS)建立小鼠溃疡性结肠炎模型。50只BALB/c小鼠随机分为正常组、模型组、酪酸梭茵组、美沙拉嗪纽、酪酸梭菌联合美沙拉嗪组。观察指标包括疾病活动指数(DAI)评分,结肠粘膜肉眼改变及病理组织学积分,采用免疫组化法检测结肠粘膜组织中HSP27的表达量,ELASA法检测血清中TNF—α的表达量。结果酪酸梭茵和美沙拉嗪都可以降低实验小鼠DAI积分,减轻肠道的组织学损伤程度;与模型组相比,酪酸梭茵组、美沙拉嗪组、酪酸梭菌联合美沙拉嗪组,结肠中HSP27的表达都上调(P〈0.05),血清中TNF-α的表达都下降(P〈0.05),其中美沙拉嗪组与酪酸梭茵组治疗效果相当(P〉0.05),酪酸梭菌联合美沙拉嗪组治疗效果最佳(P〈0.05)。结论酪酸梭菌和美沙拉嗪对急性期UC都有治疗作用,二者联合用药效果更佳,其部分机制可能与上调HSP27,抑制TNF—α的表达而减轻肠道的炎性反应有关。
