小鼠胎儿胸腺T细胞体外培养的分化和成熟

摘    要:目的 :观察 BALB/ c小鼠胎儿胸腺 T细胞体外培养的分化和成熟。方法 :采用小鼠胎儿胸腺器官体外培养体系及流式细胞术测其 T细胞 CD4、CD8和 TCR的表达。结果 :受孕 15 d胎鼠胸腺 T细胞主要为 CD4 -CD8-细胞 (96 .0 % ) ;体外培养 3d后 ,大部分分化为 CD4 + CD8+ 细胞 (6 5 .4 % ) ,小部分成熟为 CD4 + CD8-(14.3% )和 CD4 -CD8+ (11.2 % )细胞 ;培养 5 d后 ,CD4 + CD8+ 细胞 (5 9.7% )减少 ,而CD4 + CD8-(14.3% )和 CD4 -CD8+ (15 .8% )增多 ;培养后 10 d,CD4 + CD8+细胞降低到 4 7.2 % ,CD4 + CD8-和 CD4 -CD8+ 细胞分别增加到 2 0 .0 %和 2 4 .9% ,同时 TCR明显表达。结论 :应用小鼠胎儿胸腺器官培养体系进一步通过胸腺细胞的阳性和阴性选择证实其 T细胞的分化和成熟过程。

Abstract:Objective: To observe the differentiation and maturation of T cells in BABL/c mouse fetal thymus in vitro. Methods: The expressions of T cell CD4, CD8 and TCR were datermined with mouse fetal thymus organ culture system and flow cytometry. Results: The T cells of fetal thymus (day 15 of gestation) were mainly the CD4 -CD8 - cells (96 0%). The greater part of the CD4 -CD8 - cells was differentiated into the CD4 +CD8 + cells (65 4%) 3 days after thymus organ culture in vitro, and the smaller part of them was matured into the CD4 +CD8 -(14 3%)and CD4 -CD8 +(11 2%)cells. The CD4 +CD8 + cells (59 7%) decreased 5 days after culture, while the CD4 +CD8 - (14 3%) and CD4 -CD8 + (15 8%) cells increased The CD4 +CD8 + cells decreased by 47 2% 10 days after culture, while the CD4 +CD8 - and CD4 -CD8 + cells increased by 20 0% and 24 9%, respectively, meantime the TCR expressed obviously. Conclusion: The course of T cell differentiation and maturation was further demonstrated by thymocyte positive and negative selections with mouse fetal thymus organ culture system.
