摘    要:目的从抗氧化的途径探讨三七总皂甙(PNS)静脉给药对油酸致大鼠急性肺损伤的作用及其机制.方法健康雄性SD大鼠40只,用25%乌拉坦腹腔注射麻醉(6mL/kg)后随机分为3组:(1)正常对照组(n=8),不做任何处理;(2)急性肺损伤组(n=8),股静脉缓慢推注油(0.12mL/Kg);(3)三七总皂甙组(n=24),股静脉缓慢推注油酸(0.12mL/Kg),15分钟后股静脉注入三七总皂甙,又分为低剂量(25mg/kg)、中剂量(50mg/kg)和高剂量(75mg/kg).4h后断头处死大鼠,开胸取肺.HE染色光镜下观察肺组织病理形态学变化,检测肺湿干比值(W/D),生物化学方法检测肺组织匀浆一氧化氮(NO)和丙二醛(MDA)含量及髓过氧化物酶(MPO)、一氧化氮合酶(NOS)和超氧化物歧化酶(SOD)的活力.结果(1)肺组织病理学变化:急性肺损伤大鼠肺组织呈现典型的炎症病理特征,包括有肺泡壁明显增厚,肺泡和肺间质水肿、大量中性粒细胞浸润,肺泡腔缩小变形,肺泡壁破裂,肺泡塌陷实变等急性炎症损害表现,并伴有明显的充血及大量暗红色的组织梗死区.三七总皂甙组各剂量均能够明显的减轻肺炎症反应和肺的损伤程度.(2)其它指标结果显示:与正常对照组比较,急性肺损伤组肺组织W/D比值、NO、MDA含量及MPO、NOS的活力均增高(P〈0.05),而SOD活力明显下降(P〈0.05);三七总皂甙组各剂量均能降低急性肺损伤的肺组织W/D比值、MDA含量及MPO、NO、NOS的活力(P〈0.05),提高SOD活力(P〈0.05),且呈剂量依赖关系.结论三七总皂甙静脉给药能够明显的减轻油酸所致急性肺损伤的炎性反应和损伤程度,降低肺W/D比值、一氧化氮合酶(NOS)和髓过氧化物酶(MPO)的活性及丙二醛(MDA)、一氧化氮(NO)含量,提高肺组织超氧化物歧化酶(SOD)的活力,其药理作用机制可能与提高肺内抗氧化作用有关.

Abstract:Objective To explore the antioxidative effects of panax notoginseng saponins(PNS) on rats with acute lung injury induced by oleic acid.Methods 40 healthy male SD rats were randomly divided into three groups:(1) normal control group(n=8);(2) acute lung injury group(n=8),rats were injected oleic acid(0.12 mL/kg);(3) PNS intervention group(n=24),rats were injected by PNS(25 mg/kg,50 mg/kg,75 mg/kg,respectively) after oleic acid(0.12 mL/kg) injection 15 minutes.Animals of all three groups were sacrificed after 4 hours.The pathological changes,lung wet/dry(W/D) weight ratio,contents of malondialdehyde(MDA) and nitric oxide(NO),and activities of myeloperoxidase(MPO),nitric oxide synthase(NOS),and superoxide dismutase(SOD) were measured in lung tissue.Results(1) Pathological changes of acute lung injury were reduced significantly in PNS intervention groups.(2) Compared with normal control group,W/D,MPO,NO,NOS and MDA in acute lung injury group were all higher(P〈0.05),SOD was lower;W/D,MPO,NO,NOS and MDA in PNS groups were all lower than acute lung injury group(P〈0.05),SOD was higher.Conclusion These results suggest that PNS can alleviate lung inflammation,decrease the W/D,MPO,MDA,NO,NOS and increase SOD,and decrease the degree of acute lung injury in rats injecteel by oleic aeid,and that may be involved in increase of lung antioxident action.
