芦荟多糖对脑缺血大鼠p53蛋白表达的影响

摘    要:目的观察芦荟多糖(AP)对局灶性脑缺血大鼠的神经保护作用,并进一步探讨其作用机制。方法采用线栓法阻断大鼠一侧大脑中动脉(MCAO),制备局灶性脑缺血模型,观察芦荟多糖对MCAO模型大鼠脑缺血后神经功能缺损导致的行为学变化,通过免疫组化法检测其对大脑皮质细胞凋亡相关蛋白p53表达的影响,以此探讨芦荟多糖对脑缺血的神经保护作用。结果芦荟多糖能明显降低神经功能缺损评分和抑制MCAO模型大鼠大脑皮质p53蛋白表达。结论芦荟多糖对急性脑缺血具有神经保护作用,其机制可能与芦荟多糖影响凋亡相关蛋白p53的表达有关。

Effect of Aloe Polysaccharide on protein expression of p53 in rats with focal cerebral ischemia

Abstract:Objective To observe the neuro protective effects and explore the further mechanism of Aloe Polysaccharide(AP) on cerebral ischemia in rats. Methods Wistar rats with incomplete cerebral ischemia were induced by ligating the middle cerebral artery occlusion (MCAO) with fishing lines, It was detected the effects of Aloe Polysaccharide on protein expression of p53 with immuneohistochemical method. Meanwhile, It was observed the effect of AP on the changes of ethology due to nerve injury of rats with MCAO before and after treatments. Results AP can inhibit the expression of p53 protein in cerebral cortex and reduce the neurological deficit score. Conclusion Aloe Polysaccharide has protecting effect on cerebral ischemia and the mechanism of Aloe Polysaccharide against cerebral ischemia is related to affecting the related expression of p53 protein.
