日本血吸虫成虫在体外培养中DNA合成的影响因素的实验研究

摘    要:使用 ̄3H胸腺嘧啶核苷摄取作为日本血吸虫体外培养DNA合成指标的研究发现:①在体外培养不同时间,血吸虫DNA合成速度不同。在刚进入离体培养的20h内,血吸虫的DNA合成被抑制在较低的水平。随后,血吸虫雌雄虫的DNA合成迅速上升达到最高水平。在培养的1~6d,血吸虫DNA合成虽略有变化,但基本上维持在较高水平。在培养的6~12d,血吸虫DNA合成迅速降低到低水平,②雌雄虫的DNA合成不同,雌虫DNA合成较雄虫高,差别有高度显著性。③培养基中的血清浓度不同,DNA合成速度不同。当低于10%血清浓度,有随着血清浓度的增加.DNA合成也呈增加的趋势。当高于10%血清浓度,随着血清浓度的增加.DNA合成受抑制。在10%血清浓度和20%血清浓度的DNA合成的差别有显著性(P<0.05).证明10%血清浓度是培养基中最适血清浓度。最后,讨论了各种影响体外培养日本血吸虫成虫DNA合成因素的意义。

Abstract:The effects of maintenance in vitro and different concentration of newborn calf serum on DNA synthesis in adult Schistosoma japonicum cultured in vitro was investigated using the uptake of 3H thymidine as a specific marker of DNA synthesis.1) during 20 h cultivation, the specific activity of DNA synthesis was significantly despressed (in males,<0. 002; in males,P<0. 005) compared to an elevated uptake of 3H-TdR on day 1 of DNA synthesis was not significant and both sexes had a high uptake of 3H-TdA. At 12 days post-culture the specific activity of DNA synthesis was significantly depressed in comparision with to that observed on day 6 of culture(in males,P<0.01; in females, P<0.002). 2)The difference in 3H-TdR uptake between adult males and females was highly significant. 3) When the concentration of newborn calf serum was at 10%, culturing both sexes had higher specific activity of DNA synthesis than at 20% or 40%. It appears that the medium containing 10% concentration of newborn calf serum is suitable for the DNA synthesis of adult schistosomes.
