脑瘫兔血清及脑组织中髓鞘碱性蛋白含量的研究

摘    要:目的 :探讨髓鞘碱性蛋白 (MBP)能否作为脑瘫早期诊断生物学指标及反映各脑区损伤情况。方法 :出生后 3d仔兔腹腔分次注射胆红素 (总量 30 0mg/kg)制备出兔脑瘫动物模型 ,观察脑组织不同区域病理变化 ;采用酶联免疫法测定血清和脑组织中MBP含量。结果 :仔兔注药后 12h血清中MBP含量实验组较对照组明显升高(P <0 .0 5 ) ,而脑瘫仔兔与对照组比较差异无显著性。脑组织中MBP含量脑瘫仔兔海马、基底节区较对照组减少(P <0 .0 5 ) ,脑干、小脑、皮层区与对照组比较差异无显著性 (P >0 .0 5 )。结论 :血清MBP含量可作为胆红素致脑损伤早期诊断的敏感指标 ;胆红素致脑瘫兔脑海马、基底神经节区髓鞘脱失 ,但在脑干、小脑、皮层变化不明显。

Abstract:Objective:To discuss whether the contents of serum myelin-basic protein (MBP) can be used as sensitive mark to early diagnosis of brain damage induced by bilirubin. Methods: A cerebral palsy (CP) rabbit model was developed by intraperitoneal infusion of bilirubin (total 300 mg/kg) in the rabbits of 3 days old. The pathologic changes of different brain regions were observed and the contents of MBP in serum and brain tissues were detected by ELISA. Results: The contents of serum MBP in CP rabbits 12 h after administration of bilirubin were significantly higher than those in control group (P<0.05), but there was no significant difference between CP rabbits 45 days after administration of bilirubin and control group. The contents of MBP in the regions of hippocampus, basal ganglia in CP rabbits were significantly decreased as compared with those in the control group (P<0.05). Conclusion: The contents of serum MBP can be used as a sensitive mark to early diagnosis of brain damage induced by bilirubin. The number of neurons was decreased in hippocampus and basal ganglia, and the myelin was degenerated in brainstem, hippocampus, basal ganglia in the CP rabbits induced by bilirubin.
