摘    要:用RT PCR法克隆抗RSV 鼠单抗4C2 的轻、重链可变区基因, 构建了轻、重链分别表达的载体pAG4622_4C2 和pAH4604 4C2, 及轻、重链同时表达的载体pdHL4C2, 分别转染SP2/0 细胞和DHFR- CHO细胞, 检测培养上清中的嵌合抗体表达。结果在pdHL4C2 转染的CHO 细胞中检测出了可与表达RSVG/F糖蛋白的143TK 细胞结合的人 鼠嵌合抗体

Abstract:An anti RSV human mouse chimeric antibody was constructed,firstly by cloning the light and heavy chain of the mouse anti RSV monoclonal antibodies 4C2 by RT PCR,and then constructing the light chain expressing vector pAG4622 4C2,heavy chain expressing vector pAH4604 4C2,as well as the simultaneously expressing vector pdHL4C2 Transfections of SP2/0 and DHFR - CHO cells with these constructed genes were conducted respectively It was found that the human mouse chimeric antibodies that could bind with the RSV G/F glycoprotein might be detected in the supernatant of pdHL4C2 transfected CHO cells
