| 摘 要： | 目的 探讨尼卡地平体外对大鼠系膜细胞凋亡相关基因(Bcl-2／Bax)表达的影响。方法 采用多克隆抗体间接标记，流式细胞仪检测大鼠系膜细胞凋亡相关基因(Bcl-2／Bax)的蛋白表达；以β-actin为内参标。RTPCR法测定以上基因的mRNA表达；Ho33342染色测定细胞凋亡。结果 尼卡地平在蛋白及mRNA水平均下调Bcl-2基因的表达，对Bax基因的表达则无明显影响。结论 尼卡地平下调Bcl-2基因表达是引起大鼠系膜细胞凋亡的主要原因。

| Abstract: | Objective To study the mechanisms of rat mesangial cell apoptosis by Nicardipine in vitro. Method Apoostosis was assessed by Ho33342 staining;Bcl-2/Bax mRNA was detected by RT-PCR and the protein of this genes was valued by indrect-immuno staining with flow cytometer. Result Nicardipine reduced the expression of Bcl-2 gene of rat mesangial cell in mRNA and protein levels. Conclusion Down-regulation of Bcl-2 gene expression is the major mechnism of rat mesangial cell apoptosis which was induced by Nicardipine. |
