胶质瘤细胞系SHG44中肿瘤干细胞存在的可能性

摘    要:[摘要] 目的 探讨人类胶质瘤SHG44干细胞存在的可能性。方法 应用N2无血清培养基进行培养,碱性成纤维细胞生长因子和表皮生长因子刺激细胞扩增;应用免疫组织化学染色对培养的细胞及其分化的细胞进行鉴定。结果 SHG44在无血清的培养条件下细胞的形态明显改变,部分细胞聚集成球,类似神经干细胞的神经球,呈悬浮状态生长,其余细胞贴壁生长,细胞体呈球形或椭球形,伸出双极或多极的突起。细胞表达波形蛋白和巢蛋白两种神经干细胞的标志物,细胞表达胶质纤维酸性蛋白;大部分细胞也表达神经元的标志物神经元特异性烯醇化酶、神经微丝蛋白200和βⅢ管蛋白。结论 体外的培养条件下,胶质瘤细胞系SHG44可以同时表达神经元和星形胶质细胞表型,具有干细胞的性质。

The Existence of Glioma Stem Cells in Human SHG44 glioma cell line
Abstract:[Abstract] Objectives To explore the possibility of glioma stem cells (GSCs) existence in SHG44 glioma cell line. Methods SHG44 were cultured in N2 medium with basic fibroblast growth factor (bFGF) and epidermal growth factor (EGF) for some times, and they were identified by immunocytochemistry. Results SHG44 glioma cells were cultured in neural stem cell medium, and they could formed clusters in suspension, some adherent cells were spherical or ellipsoid shape extending bipolar or tri-polar processes, they could be cultured and passaged in vitro for 3-5 weeks, but they could not proliferate for long-term in this medium. The majorities of the cells expressed vimentin and nestin, which were the markers for neural stem cells. They could express glial fibrillary acidic protein, beta Ⅲ-tubulin, neuron specific enolase (NSE), neurofilament 200 (NF200) simultaneously, the markers for neurons and astrocytes. Conclusions SHG44 Human glioma cells could express the markers for neurons and glia, and they possess the characteristics for stem cells.
