摘    要:目的:研究将RGD肽结合至聚乳酸基复合材料-聚乙交脂丙交脂与羟基磷灰石的复合物,形成的新型RGD-PLGA/HA(RGD peptide modified poly lactic-glycolic acid/hydroxyapatite)膜材料的细胞亲和性。方法:以RGD-PLGA/HA膜和作为对照的未结合RGD肽的PLGA/HA材料为实验对象,取第二代兔骨髓基质细胞(bone marrow stromal cells,BMSCs)接种到材料上,培养4、12、24h后,细胞计数仪计数定量检测粘附的细胞数,得到粘附率;培养4、24、96h后环境扫描电镜比较粘附细胞的形态和质量。结果:实验组材料与BMSCs复合培养4、12h后,测定细胞粘附率分别为(40.5±1)%、(46.5±0.5)%,均高于同期对照组(P<0.05);24h后实验组材料的细胞粘附率(57.3±0.5)%,也较对照组(47.5±0.5)%高(P<0.01);环境扫描电镜观察BMSCs与实验组材料的细胞形态和质量明显优于对照组。结论:与PLGA/HA相比,新型RGD-PLGA/HA膜材料能提高其对BMSCs的亲和性。

Abstract:Objective: To evaluate the cellular affinity of RGD peptide modified PLGA/HA(poly lactic coglycolic acid/ hydroxyapatite) film. Methods: The rabbit bone marrow stromal cells (BMSCs) of generation 2 were seeded onto the biomaterials (RGD-PLGA/HA and PLGA/HA) and incubated for 4?12 and 24 hours at standard condition. At each time point the number of adhesive cells was counted. The environmental scanning electron microscope was used to take photo of adhesive cells cultured for 4, 24 and 96 hours, to evaluate the quality of cell adhesion. Results: At 4 and 12 hours, the efficiency of cell adhesion on RGD-PLGA/HA was higher than that on unmodified PLGA/HA. At 24 hours the quality of cell adhesion on RGD-PLGA/HA was better. Conclusion: BMSCs adhesion to RGD-PLGA/HA can be enhanced by modifying the PLGA/HA film with RGD peptide.
