免疫组织化学染色检测食管组织MGMT和P53蛋白的表达及其相关性

摘    要:目的探讨MGMT在食管上皮单纯增生、非典型增生、原位癌和浸润癌组织的表达及其与P53的关系。方法应用免疫组化(EnVision二步法)检测21例正常上皮、39例单纯增生上皮、17例非典型增生、12例原位癌以及140例浸润癌组织中MGMT及P53蛋白的表达及其相关性。结果MGMT蛋白表达强度在原位癌和非典型增生上皮最高,浸润癌组织次之,单纯增生上皮和正常上皮表达较低,统计学分析其差别有统计学意义(F=3.022,p<0.05)。P53在正常上皮不表达,从单纯增生上皮→非典型增生上皮表达逐渐增强,在原位癌和浸润癌组织中表达强度最强。统计学分析各组表达强度存在差异(F=16.18,p<0.01)。MGMT和P53的相关分析结果显示,在癌组织MGMT与P53蛋白之间的表达呈负的相关关系,(rs=-0.415,p<0.01)。结论MGMT和P53蛋白在食管组织癌变的过程中的表达改变可能与食管癌发生有关。

Abstract:Objective Expression of MGMT and mtP53 was detected immunohistochemically in esophageal cancer tissue to found out the relation between them in esophageal carcinogenesis. Methods P53 and MGMT were observed by Envision immunohistochemistry method in 21 cases with normal epithelium,39 with hyperplasia, 17 with dysplasia,12 with carcinoma in situ and 140 with invasive carcinoma. Results There was significant difference of the expression intensity of MGMT among the various categories of the esophageal tissue ( F = 3. 022, p 〈 0. 05). There was significant difference of expression positive rate(X^2 =32. 043, p 〈 0. 01 ) and the expression intensity of P53 among the various categories of the esophageal tissue also had difference (F=16.18, p〈0. 01 ). The expression of MGMT was negatively correlated with P53 in carcinoma tissue (X^2=-0. 415, p〈0.01). Condusion The changes of the expression of MGMT and P53 may be correlated with the genesis of esophagus cancer.
