单侧输尿管梗阻大鼠肾小管间质MCP-1的表达及其意义

摘    要:目的:探讨肾间质纤维化中单核细胞趋化因子-1(MCP-1)的表达及其与肾间质巨噬细胞浸润的关系。方法:60只SD雌性大鼠随机分成假手术(SOR)组、单侧输尿管梗阻(UUO)组,于术后第1,4,7,10,14 d分别处死各组大鼠。用HE和MASSON染色法观察UUO大鼠术后不同时间点肾脏病理改变,用免疫组化法检测肾小管间质MCP-1表达和巨噬细胞浸润。结果:UUO组肾小管-间质MCP-1,单核巨噬细胞抗原ED-1表达较SOR组明显增多(P<0.05),MCP-1表达与巨噬细胞浸润呈正相关(r =0.865,0.928,0.858,0.893,0.873,均P<0.05);MCP-1表达和巨噬细胞浸润分别与肾间质胶原相对面积呈正相关(分别为r =0.856,0.896,0.686,0.820,0.867和r =0.942,0.898,0.920,0.914,0.829,均P<0.05)。 结论:随UUO大鼠病程进展,肾小管间质MCP-1表达增多,可能在肾间质纤维化中起重要作用。MCP-1可能通过介导巨噬细胞浸润参与肾间质纤维化,MCP-1可成为治疗肾间质纤维化的新靶点。

Abstract:Objective To investigate the renal expression of monocyte chemoattractant protein-1(MCP-1) in rat with unilateral ureteral obstruction(UUO) and its association with macrophages infiltration in renal interstitium. Methods Sixty female Sprague-Dawley rats were randomized into UUO and sham- operation(SOR)groups.The kidneys were harversted at 1,4,7,10 and 14 d after the operation. Immunohistochemistry was used on the renal tissue for MCP-1 and ED-1 which was a marker of macrophage. Histological changes were also observed by HE and Masson staining.Results Compared with the SOR group,there was a significant increase in MCP-1,ED-1 expression in the UUO group(P<0.05). The expression intensity of MCP-1 was correlated with the interstitial macrophages accumulation and interstitial fibrosis (r=0.865,0.928,0.858, 0.893, and 0.873;r=0.856,0.896, 0.686, 0.820, and 0.867,respectively; P<0.05). Interstitial macrophage infiltration was correlated with the grade of interstitial fibrosis too(r =0.942,0.898,0.920,0.914,and 0.829,P<0.05). Conclusion Elevated MCP-1 expression might be responsible for the macrophages infiltration and interstitial fibrosis in the obstructed kidney.
