胃癌中COX-2 基因的表达及其与NF-κB的相关性

目的:研究胃癌中环氧合酶2(COX-2)的表达情况和生物学意义,以及与核因子κB(NF-κB)的相关性。方法:选择胃癌及癌旁正常组织41例,对COX-2、NF-κB进行RT-PCR及Western blot检测。结果:41例胃癌标本中有28例COX-2阳性,26例NF-κB阳性,COX-2和NF-κB表达呈正相关。COX-2和NF-κB基因表达与年龄、性别、大小、组织学类型无关,但与浸润深度、临床分期和肿瘤的淋巴结转移具有显著的相关性(P<0.05)。结论:COX-2过表达在肿瘤侵袭过程中发挥着重要作用,而NF-κB可能通过调节COX-2的表达在胃癌细胞增殖过程中起重要作用。

Expression of Cyclooxygenase in Gastric Carcinoma and Its Correlation with NF-kappaB

Objective: To explore the biological significances of cyclooxygenase(COX) in patients with gastric cancer and its correlation with NF-κB.Methods: Forty-one cases of gastric carcinomas and normal tissues were detected for COX-2 and NF-κB with RT-PCR and Western blot.Results: Of the 41 cases,28 showed positive COX-2 expression and 26 showed positive NF-κB expression.The expression of COX-2 was positiveily correlated with those of NF-κB(P< 0.05).COX-2 and NF-κB expression was not correlated with such clinicopathological parameters as age,sex,size and histological types.However,there was a positive relationship between COX-2 gene expression and depth of invasion or lymph node metastasis.Conclusion: The study demonstrated that the overexpression of COX-2 gene may participate in the oncogenesis and progression of gastric carcinoma.NF-κB plays an important role in the process of invasion and metastasis of tumor cells by inducing high expression of COX-2.
