低氧促进人脐带血间充质干细胞向神经细胞分化

摘    要:目的探讨低氧环境对脐带血间充质干细胞诱导分化成神经细胞的促进作用。方法收集4例志愿者脐带血,分离获得间充干细胞,体外扩增培养。研究分为常氧组、常氧诱导组、低氧组、低氧诱导组。研究低氧诱导培养的脐带血间充质干细胞组与对照组比较在细胞生物学特征、诱导分化能力等方面的差异。结果在5%含氧量条件下细胞的增生能力不同程度的提高;在诱导成神经细胞的过程中,低氧诱导的间充质干细胞向神经细胞的分化能力增强。神经细胞表面标记Nestin、NF-M表达阳性。与常氧诱导的脐带血间充质干细胞组比具有显著性差异(P〈0.05)。结论低氧条件可以提高脐带血间充质干细胞向神经细胞分化的产率。

Abstract:Objective To investigate human umbilical cord blood mesenchymal stem cells induced and differ- entiated into neural cells under hypoxic environment. Methods To collect four cases of cord blood, stem cells ob- tainedand cultured in vitro. The cells were divided into normal oxygen group, induction group in normal oxygen, hy- poxia group, and hypoxia - induced group. The experiment was to detect the biological characteristics and the differ- entiation - ability of hypoxia - induced mesenchymal stem cell group compared with the control group. Result The proliferative capacity of umbilical cord blood mesenchymal stem ceils was improved fewer than 5 % of oxygen. The capacity of the hypoxia - induced mesenchymal stem ceils differentiated to nerve cells was increased. Nestin and Neurofilament - M of Nerve cell surface marker were positive expression, here is significant difference between hy- poxia - induced group and normal oxygen - induced mesenchymal stem cell group ( P 〈 0.05 ). Conclusion Hy- poxic conditions can simulate human umbilical cord blood mesenchymal stem ceils differentiate into neural cells and that may provide some theoretical basis for the development and application of neural tissue engineering.
