改进SP法检测热休克蛋白

摘    要:目的 :探索避免常规SP法检测热休克蛋白 (HSP)时出现假阳性和消除非特异性反应的方法。方法 :心肌组织石蜡切片 ,采用改进的SP法染色 ,即在常规SP法基础上“一抗”倍比稀释 ,双蒸水稀释H2 O2阻断内源性过氧化物酶等染色操作。结果 :改进的SP法染HSP阳性产物呈棕黄色细颗粒状 ,在细胞浆中弥漫分布 ,背景十分清亮。结论 :改进后的SP法更适用于HSP的检测

Abstract:Establishing the improved SP methods to avoid the phe nomenon of false positine and eliminate the non specificity reaction during the test of HSP. Method: Using the wax slices of mycocardium tissue and the impro ved SP dyeing methods with "the first antibody" serial dilutions the routine t rypsim to digest the repared antigen and 3% H 2O 2 to prevent the activity of endoperoxidase.Results: These improvements can successfully avoid the phenomeno n of false positive resuct the positive products of HSP dyeing present in fine b rown particle which spread in cytoplasma and the dyeing background is very clear .Conclusion: The improved sp methods can be applied to the clinic al HSP testing.
