大鼠肠缺血再灌注后肠黏膜细胞凋亡和内源性一氧化氮的观察

摘    要:目的 通过观察大鼠肠缺血再灌注后肠黏膜细胞凋亡及血清一氧化氮(NO)浓度改变的情况,旨在为临床提供检测肠缺血再灌注损伤的指标。方法 成年雄性SD大鼠,夹闭肠系膜上动脉1h,再开放2h制成肠缺血再灌注的7只动物模型作为实验组;正常假手术大鼠5只为对照组。测定缺血前后及再灌注后的血清NO浓度、再灌注后肠黏膜细胞TUNEL染色的阳性细胞数,采用免疫组织化学ABC法检测半胱氨酸天门冬氨酸特异蛋白酶(Caspase—3)的表达水平。结果 实验组在缺血前清NO浓度与对照组的差异无显著性;缺血后及再灌注后明显下降(P<0.05),与对照组比较,差异有显著性。缺血再灌注后实验组肠黏膜细胞的凋亡增加,TUNEL染色阳性细胞数及Caspase—3平均灰度值均显著高于对照组(P<0.01)。结论 大鼠肠缺血再灌注后血清NO浓度下降,肠黏膜细胞凋亡增加,可作为肠缺血再灌注的损伤指标,适用于临床辅助诊断并为治疗提供参考。

Abstract:Objective To observe the changes of serum nitric oxide and mucosal cell apoptosis after intestinal ischemia reperfusion in rats and to provide the detective markers in clinic.Methods Healthy S D rats were anesthetized and divided into 2 groups: Group A as control, by sham operation, ( n =5); Group B with occlusion of superior mesenteric artery for 1 hour followed by 2 hours reperfusion, ( n =7). Determining the serum nitric oxide before and after intestinal ischemia reperfusion, also the positive cell number of TUNEL and concentration of caspase 3 in mucosal cells. Results Before intestinal ischemia reperfusion, the serum nitric oxide concentration of the two groups were similar,in group B showed obviously decrease after reperfusion( P <0.05) with increase of mucosal cell apoptosis increased, including positive cell number of TUNEL and caspase 3 which were both higher than those in Group A ( P <0.01).Conclusion Decrease of concentration of serum nitric oxide with increase of mucosal cell apoptosis could be taken as the markers of intestinal ischemia reperfusion injury and helpfulness for the diagnosis and management in rats.
