| 摘 要： | 目的 探讨2-甲氧基雌二醇(2-ME)对体外培养的U87胶质瘤细胞增生、凋亡及细胞周期的影响.方法 将不同浓度的2-ME分别作用于U87细胞,采用倒置显微镜观察细胞形态学变化;四甲基偶氮哩蓝(MTT)法检测2-ME对U87细胞增生的抑制作用;流式细胞术分析2-ME对U87细胞凋亡及细胞周期的影响.结果 1～15μmol/L浓度的2-ME在24、36、48及72 h均可明显抑制U87细胞增生并诱导其凋亡.并在一定范围内呈时间和剂量依赖性;各亚组与对照组差异具有统计学意义(P<0.05).U87细胞经2-ME作用后G2/M期细胞增加,而G0/G1期和S期细胞减少.结论 2-ME可明显抑制U87胶质瘤细胞增生并诱导其凋亡,在一定范围内呈时间和剂量依赖性.

| Abstract: | Objective To explore the effects of 2-methoxyestradiol(2-ME) on proliferation,apoptosis and cell cycle of U87 cells in vitro. Methods U87 cells were treated with 2-ME of different concentrations and time in vitro,morphological changes of apoptotic cells were observed under microscope;cell proliferation inhibition was detected by MTT colorimetric assay;cell cycle arrest and apoptosis rate were measured by flow cytometry(FCM).
