小鼠螺旋神经节发育过程中胰岛素样生长因子-1表达的变化

摘    要:目的观察胰岛素样生长因子-1(IGF-1)在小鼠螺旋神经节发育过程中的表达和分布特点,探讨其生物学特性及意义。方法制作耳蜗组织切片,采用多重荧光免疫组织化学染色方法,激光共聚焦显微镜观察小鼠出生后12 d的IGF-1在耳蜗螺旋神经节的分布和定位。应用Western blot及Real-time PCR法检测新生小鼠螺旋神经节在出生后不同时期IGF-1蛋白及IGF-1 mRNA的表达水平。结果 IGF-1在小鼠耳蜗发育期中在螺旋神经节均有表达,表达在整个细胞体上。小鼠耳蜗螺旋神经节的IGF-1蛋白及IGF-1 mRNA表达水平随着出生年龄增加而逐渐减少。结论 IGF-1在小鼠耳蜗发育期中在耳蜗螺旋神经节均有表达。表达量随着出生年龄增加而逐渐减少。推测其可能通过旁分泌和自分泌方式在局部发挥其功能,在内耳细胞增殖中可能起重要信号转导作用。

Abstract:Purpose To study the expression and distribution of Insulin-like Growth Factor(IGF-1) in the spiral ganglion cells of the normal developing mice,and to elucidate the function and biological characteristics of IGF-1.Methods The expression and distribution of IGF-1 of the spiral ganglion cells during normal developing mice was observed by multiplicitas fluorescence immunohistochemistry with laser scanning confocal microscope.IGF-1 protein and m RNA was examed by Western blot and RT-PCR.Results In the spiral ganglion cells of cochlea,multifluorescence immunohistochemically showed that IGF-1 was widely distributed in the cell body of the spiral ganglion cells.The protein and mRNA expression of IGF-1 in the spiral ganglion cells and the quantity of expression decreased with aging.Conclusion There was expressions of IGF-1 in the spiral ganglion cells in normal developing mice.The expressions are related to ages,when the age increased,the expressions will decrease.IGF-1 may play an important role in signal transduction during inner ear cell proliferation as a paracrine and autocrine manner.
