兔心肌中次黄嘌呤纯化工艺的优化

摘    要:目的探索从兔心肌中提取次黄嘌呤的最佳工艺。方法比较三氟乙酸法、酶解法、酸碱法和醇沉法提取的次黄嘌呤的产率和纯度,得到优化制备工艺。结果利用醇沉法,向细胞充分破碎后的匀浆液中加入4倍体积的纯水,超声提取,向提取液中加入乙醇至浓度60%,置60℃水浴反应120 min,提取的次黄嘌呤产率为0.83 mg/g,纯度为82.4%。结论经过纯化工艺比较,醇沉法提取的次黄嘌呤的含量和纯度优于其他方法。

Abstract:Purpose To investigate the content of the different extraction methods of preparation of rabbit myocardial tissue fluid hypoxanthine,to determine the hypoxanthine optimal purification process.Methods Determination of the Trifluoroacetic acid method,enzymatic,ethanol and acid-base extraction hypoxanthine content and purity.Results The tissue fluid too the cells fully broken in four times the volume of pure water,adding ethanol to a concentration of 60% water bath at 60 ℃ for 2 h,extracted hypoxanthine content of 1.410 mg/mL.Conclusion The purification process,compared to the content and purity of the alcohol precipitation extracted hypoxanthine is superior to other methods.
