卵巢上皮性癌组织中TGFBI基因启动子甲基化及VEGF检测

摘    要:目的:检测卵巢上皮性癌组织中TGFBI基因启动子的甲基化状态及VEGF的表达,探讨二者的关系。方法:应用甲基化特异性PCR法(MSP)和免疫组化SP法分别检测20例正常卵巢组织、20例卵巢良性上皮性肿瘤组织及30例卵巢上皮性癌组织中TGFBI基因启动子的甲基化状态及VEGF蛋白的表达。结果:3种组织中TGFBI基因启动子的甲基化率及VEGF蛋白的阳性表达率差异均有统计学意义(χ2=34.886和26.298,P<0.001),卵巢上皮性癌组织中TGFBI基因的甲基化率及VEGF蛋白的阳性表达率均高于正常卵巢组织和卵巢良性上皮性肿瘤组织。卵巢上皮性癌组织中TGFBI基因启动子甲基化与VEGF的表达有关联(χ2=13.976,rP=0.516,P<0.001)。结论:TGFBI基因启动子在卵巢上皮性癌组织中发生超甲基化,可能诱导血管生成,促进卵巢上皮性癌的发生发展。

Abstract:Aim:To investigate the promoter hypermethylation of TGFBI and its relationship with angiogenesis in epithelial ovarian carcinoma.Methods:Methylation-specific PCR and immunohistochemical SP method were used to separately detect the promoter hypermethylation status of TGFBI and the expression of VEGF protein in 20 cases of normal ovarian,20 cases of benign epithelial ovarian tumor and 30 cases of epithelial ovarian cancer tissues.Results:The methylation rate of TGFBI and the positive expression rate of VEGF among the 3 groups had significant differences(χ2 = 34.886 and 26.298,P < 0.001) ;the rate of TGFBI methylation and the positive expression rate of VEGF in epithelial ovarian cancer tissue were higher than those in normal and benign epithelial ovarian tumor tissue(P < 0.017).The promoter hypermethylation of TGFBI was associated with the expression of VEGF in epithelial ovarian cancer tissue(χ2 = 13.976,rP = 0.516,P < 0.001).Conclusion:TGFBI occurs methylation frequently in epithelial ovarian cancer tissues,which might induce the angiogenesis and contribute to the carcinogenesis of epithelial ovarian cancer.
