5-氨基乙酰丙酸光动力治疗裸小鼠胃移植癌的实验研究

摘    要:目的 探讨以 5 氨基乙酰丙酸 (5 ALA)作为光敏剂对胃癌进行光动力治疗 (ALA PDT)的疗效 ,为胃癌的综合治疗寻找有效的手段。方法 接种胃癌细胞MGC 80 3于裸小鼠皮下 ,建立胃移植癌模型 ,以 5 ALA(10 0mg/kg体重 )作为光敏剂 ,采用瘤内局部注射和口服两种不同方式给药 ,继以波长 63 0nm的激光照射肿瘤局部 ,观察光动力治疗后肿瘤的生长速度和组织形态学变化。结果 接受光动力治疗的两组肿瘤生长速度明显减慢 ,肿瘤表面可见坏死 ,组织学检查见肿瘤组织有广泛坏死 ,两种给药途径 ,其疗效差异无显著性 (P >0 .0 5 )。结论  5 ALA光动力治疗对胃癌有杀伤作用 ,这将为胃癌的综合治疗提供有效的手段。

Abstract:Objective To investigate the therapeutic effect of photodynamic therapy using 5-aminolaevulinic acid (5-ALA) on gastric cancer.Methods A total of 17 nude mice with thymic aplasia were transplanted with human gastric carcinoma (MGC-803) and treated with laser light at 635 nm (light dose 150 J/cm 2,fluence rate 1 000 mW/cm 2).5-ALA (100 mg/kg body weight administered orally and intratumoral injection) was used as the photosensitizer.Five h after administration of 5-ALA,the animals were irradiated with Diomed PDT 630,which emit light with a wavelength of 630 nm.The tumor volume and weight were measured,and its morphologic changes were studied.Results The tumor growth rate was significantly reduced after ALA-PDT,and between the two treatment groups the growth rate had no significant difference.Conclusion 5-ALA-PDT could induce injury to gastric cancer,which may provide a new approach to the treatment of gastric cancer.
