角质细胞生长因子对无排卵功血子宫内膜上皮细胞PCNA、ER和PR表达的影响

摘    要:目的 观察角质细胞生长因子(keratinocyte growth factor,KGF)对离体培养的无排卵型功血子宫内膜上皮细胞EEC中增殖细胞核抗原PCNA、雌激素受体ER和孕激素受体PR表达的影响.方法 体外培养无排卵型功血子宫内膜上皮细胞,加入不同浓度的KGF作用后,免疫组化法检测PCNA、ER和PR的表达变化,RT-PCR检测ER和PR mRNA表达的变化.结果 免疫组化染色结果,PCNA蛋白表达位于细胞核,ER和PR蛋白表达主要位于细胞核内,少数可见细胞质着色;50 ng/ml KGF作用后,正常组和功血组PCNA表达分别较作用前的对照组显著增高,ER和PR表达分别较作用前的对照组显著增高(P<0.05);PCNA和ER表达的增加幅度在正常组和功血组之间无显著差异(P>0.05);而功血组PR表达的增加幅度显著高于正常组(P<0.05);50.ng/ml KGF作用后,RT-PCR方法检测正常组和功血组ER和PR的mRNA表达,分别较作用前的对照组显著增高(P<0.05).结论 KGF对体外培养的无排卵型功血和正常的子宫内膜上皮细胞均有增加PCNA、ER和PR表达的作用,但增加程度有差异,提示两组的子宫内膜上皮细胞生长均受KGF的影响,但无排卵功血可能存在与正常的子宫内膜上皮细胞不同的生长机制.
