加味小青龙汤对内毒素致急性损伤大鼠肺组织TLR4 mRNA表达的影响

摘    要:目的:探讨加味小青龙汤对脂多糖(LPS)诱导的急性肺损伤(ALI)大鼠肺组织Toll样受体4(TLR4)mRNA表达的影响。方法:将72只雄性Wistar大鼠随机分为:对照组、模型组、地塞米松组和加味小青龙汤(小、中、大剂量)组,每组再分为4h和8h两个亚组,每个亚组6只。尾静脉注射LPS 10mg/kg制备大鼠ALI模型。地塞米松组和加味小青龙汤(小、中、大剂量)组分别于造模前灌胃地塞米松和加味小青龙汤,2次/d,3d。采用实时荧光定量聚合酶链反应(PCR)检测TLR4 mRNA的表达,并于光镜下观察大鼠肺组织病理变化。结果:与模型组相比,在8h组中,加味小青龙汤(小、大剂量)组的TLR4 mRNA表达均明显降低(P<0.01)。病理学观察,模型组大鼠肺组织出现大片出血及坏死,各治疗组大鼠肺组织病理改变明显轻于模型组,肺细支气管偶见炎症改变,水肿较轻,中性粒细胞(PMN)和红细胞渗出较少。结论:加味小青龙汤能减轻内毒素致ALI大鼠肺组织损伤,对肺损伤有保护作用,其机制可能与其能降低内毒素致ALI大鼠肺组织TLR4 mRNA的表达有关。
