制附子总碱的急性毒性及对过敏性鼻炎豚鼠鼻黏膜和组胺的影响

摘    要:目的:探讨制附子总碱对过敏性鼻炎(AR)豚鼠行为学指标、鼻黏膜病理形态和血液组胺含量的影响,同时测定制附子总碱的半数致死量(LD50),对其临床应用进行安全性评价。方法:40只FMMU豚鼠按体质量随机分为正常对照组、AR模型组、辛芩颗粒组、制附子总碱组,每组10只。采用卵清蛋白全身致敏与局部攻击方法制作AR豚鼠模型,正常对照组和AR模型组给予生理盐水,辛芩颗粒组每天给予辛芩颗粒4.65g/kg,制附子总碱组每天给予总碱提取物按生药量计1.13g/kg,观察豚鼠行为学指标和鼻黏膜病理形态的变化,测定血液组胺含量;对制附子总碱进行小鼠急性毒性实验。结果:制附子总碱组鼻痒、喷嚏、清涕等行为学指标及鼻黏膜炎症程度均有明显改善,血液组胺含量明显降低;制附子总碱的LD50为56.3g/kg制附子。结论:制附子总碱在临床用药方面是安全的;制附子总碱能有效缓解过敏性鼻炎的症状,降低血液组胺含量,改善鼻黏膜炎症的局部浸润,达到治疗过敏性鼻炎的目的。
