光化学诱导大鼠脑缺血的定量分析

摘    要:目的研究大鼠脑缺血后损伤体积和水含量的变化。方法给大鼠尾静脉注射孟加拉玫瑰红,经颅骨用激光黄色连续光束照射鼠脑15m in产生部位、大小固定的梗死灶,对梗死灶进行定量分析。先行独立分组,在脑缺血后选择1、4、24、72、168h共5个不同时间点,检测损伤体积的变化和脑组织的水含量。结果诱导缺血后在1~24h内损伤体积迅速扩大135.2%,而在24~168h内则缩小,168h时检测发现损伤较1h时缺血体积大的多,而与4h时差不多。结论组织损害的最大程度在该模型中是在前4h后达到最大,而4 h损伤的扩大是与脑的含水量有密切关系。

Abstract:Objective To study the change in the volume of the lesion and brain water content.Methods Rose Bengal(in a concentration of 7.5mg/ml of saline) was injected through tail vein in rats.Infarction of consistent size and location were produced by irradiating the brain with yellow light through the intact skull for 15min following the systemic injection of rose Bengal.We made quantitation analysis of photochemically induced focal cerebral ischemia by yellow laser in the rat.We examined the change in the volume of the lesion and brain water content,in separtate groups of rats,at different times points(1,4,24,72,and 168 h)after the induction of the ischemic lesion.Results The volume of ischemic damage increased rapidly between 1 and 24 h after the ischemic and decreased between 24 and 168 h.The lesion at 168 h was significantly larger than that following 1h of ischemia and similar to that obtained at 4 h.Conclusion The maximum extent of tissue dagmage happens within the first 4h in this model.The enlargement of the lesion after 4 h correlates closely with changes in brain water content.
