羌活水提物对迟发型变态反应及炎症反应的影响及其机制

摘    要:目的:探讨羌活水提物对迟发型变态反应及炎症反应的影响及其机理。方法:考察羌活水提物对2,4,6-三硝基氯苯(PC1)致迟发型变态反应(PC1-DTH)机制诱导的肝损伤、二甲苯诱导的耳肿胀及酵母多糖诱导的腹腔炎症等的影响;考虑羌活水提物对人Jurkat T淋巴瘤细胞分泌基质金属蛋白酶及迁移功能的影响。结果:羌活水提物诱导相和效应相给药均显著地抑制了PC1-DTH诱导的肝损伤。对二甲苯诱导的耳肿胀无影响,但可显著抑制酵母多糖诱导腹腔白细胞游出。羌活水提物对胶原诱导的Jurkat细胞分泌基质金属蛋白酶及迁移功能亦有显著的抑制作用。结论:羌活水提物抑制迟发型变态反应及炎症反应的作用可能与下调基质金属蛋白酶从而抑制白细胞的迁移功能有关。

Abstract:AIM:To examine the effects of aqueous extract from Rhizoma Notopterygii (RN ext) on the delayed type hypersensitivity (DTH) and inflammatory reactions and its mechanisms. METHOD:Liver injury induced by DTH to picryl chloride (PCl), xylene induced ear swelling and zymosan induced peritoneal inflammation were used. The assays for the activity of matrix metalloproteinase (MMP) in human Jurkat T leukemia cells and for the cell migration were also used. RESULT:RN ext significantly inhibited the DTH induced liver injury in both induction and effector phases. The extract did not inhibit the xylene induced ear swelling but significantly reduced the leukocyte recruitment induced by zymosan. RN ext also inhibited the MMP activity in Jurkat cells as well as the migration of the cells. CONCLUSION:The inhibition on the migration of leukocytes via down regulating MMP activity might be involved in the anti DTH and anti inflammatory activities of RN ext.
