摘    要: 目的:为评价草豆蔻的质量,建立其中山姜素和小豆蔻明的含量测定方法。方法:高效液相色谱法,用Shim Pack CLC-ODS柱(5μm,150mm×6mm)以甲醇-水(80∶20)作流动相,流速1ml·min-1,检测波长为300nm,外标法定量。结果:山姜素、小豆蔻明进样量在0.05μg~0.26μg范围内,进样量与峰高呈线性关系(r分别为0.9996,0.9998)。山姜素平均加样回收率为101.9%,RSD=0.18%;小豆蔻明平均加样回收率为100.0%,RSD=3.6%,n=5。测定5批草豆蔻样品,山姜素含量0.50%~1.45%,小豆蔻明含量0.07%~0.72%;草豆蔻果皮、茎叶、根茎中此2种成分的含量均在0.2%以下。结论:方法简便、可靠,可用于生药分析。

Abstract:OBJECTIVE:To establish the analysis method for alpinetin and cardamomin and evaluate the quality of Cao doukou (Alpinia katsumadai). METHOD: A HPLC method was deueloped by using a Shim Pack CLC ODS column(150mm×6mm). A mixture of methanol-water (80∶20) was used as the mobile phase with a flow rate of 1 ml per minute and the detection wavelength was set at 300nm. The contents were calculated by external-standard method. RESULTS:The standard curves for alpinetin and cardamomin were linear over the range of injected amount of 0.05μg~0.26μg(r=0.9996 and 0.9998, respectively). The average recoveries of alpinetin and cardamomin were 101.9%(n=5, RSD=0.18%)and 100.0%(RSD=3.6%), respectively. Results of analysis of 5 samples show the contents of alpinetin and cardamomin in the semens of Alpinia katsumadai were 0.50%~1.45% and 0.07%~0.72% respectively and their contents in the pericarp, stem leaves and rhizome were less than 0.2%. CONCLUSION:The mothod is convenient, reliable and can be used for analysis of crude drugs.
