摘    要:目的:探讨六味地黄汤活性部位 CA-4对小鼠 B_(16)黑色素瘤~(31)P 磁共振波谱(~(31)P MRS)的影响。方法:对照组给予生理盐水,给药组于接种 B_(16)黑色素瘤24 h 后按125 mg/kg 体重灌胃给药,连续给药11天,于药后7、9和12天测定~(31)P MRS。结果:CA-4可以有效抑制肿瘤生长,肿瘤湿重显著降低,同时使~(31)P MRS参数表现出显著性变化:给药组的磷酸肌酸/无机磷(PCr/Pi)和 ATP/Pi 显著高于对照组,而磷酸单脂(PME)/ATP 显著低于对照组(P<0.05)。提示肿瘤组织的生物能态改善,磷脂代谢受到抑制。结论:核磁共振技术作为一种无损伤的测试技术,可望成为一种抗肿瘤药物活性评价及药理作用研究的新的药效学指标,同时对于从能量代谢角度研究抗肿瘤药物作用机理也是有效的技术方法。

Effect of CA-4,an Active Fraction from Liuwei Dihuang Decoction,on ~(31)p MRS of Melanoma B_(16) in Mice

Abstract:To study the effect of CA-4,an active fraction from Liuwei Dihuang Decoction,on ~(31)p magnetic resonance spectroscopy (MRS)of melanoma B_(16) in mice.Methods:C-57 mice were inoculated with melanoma B_(16),24 hrs after the inoculation CA-4 was given (125 mg/kg)to the treated group,and to the control group normal saline was given instead,the treatment lasted for 11 days and ~(31)p MRS was measured at 7 th,9 th and 12 th day.Results:Liuwei Dihuang Decoction CA-4 could effectively inhibit the tumor growth,the tumor wet weight decreased significantly.Meanwhile the parameters of ~(31)p MRS significantly changed after CA-4 treatment,PCr/Pi and ATP/Pi ratios were significantly higher and PME/ATP lower in the treated group than those in the controt group.Conclusion:As a non-invasive method,~(31)P MRS was promised to be a new pharmacodynamic index in evaluating anticancer drugs and in studying phar- macological actions,and may be an effective method for studying the mechanism of anticancer drug viewing from energetic metabolism.
