| 摘 要： | 目的:探讨大黄素在抗肺纤维化中的作用。方法:采用体外细胞培养、免疫组化、透射电镜和同位素检测等技术,比较大黄素作用下人胚肺成纤维细胞(LF)的增殖和其分泌的白细胞介素(IL)-6的水平,检测大黄素作用下人胚 LF 电镜下的形态学特征。结果:(1)大黄素能够抑制人胚 LF 的增殖,并与其浓度、作用时间有关,大黄素高浓度组的抑制效果明显优于低浓度组,并随着时间延长其作用也明显增强;(2)在 IL-1诱导时,大黄素可以降低人胚 LF 分泌 IL-6的水平;(3)大黄素组人胚 LF 形态上与对照组比较差异无显著性。结论:大黄素具有抑制人胚 LF 增殖和抑制 IL-1刺激人胚 LF 分泌 IL-6作用。

| Abstract: | Objective:To explore the effect of emodin in anti-lung fibrosis (LF).Methods:Using extracorporeal cell culture,immunohistoche- mistry,transmission electronmicroscope (TEM) and radioisotope technology,in comparing the proliferation of human embryonic LF un- der the emodin action and the production of IL-6 secretory level,also the human embryonic LF cell proliferation cycle and the morpho- logical characteristics under TEM was investigated.Results:(1) Inhibitory effect on human embryonic LF with emodin was in a dose- dependent manner;(2)The emodin inhibited human embryonic LF to secrete IL-6 after stimulated by IL-1;(3)Emodin did not markedly change the human embryonic LF uhrastructure in emodin group under TEM examination.Conclusion:Emodin inhibited the proliferation of human embryonic LF and secretion of IL-6 induced by IL-1,it delayed the progress of cell cycle,and TEM did not show the change of cell's ultrastructure. |
