大鼠实验性脾虚证结肠电-机械活动及其与一氧化氮合酶的关系

摘    要:目的:研究脾虚证大鼠结肠运动、电活动、体表结肠电活动和结肠神经型一氧化氮合酶(nNOS)改变及四君子汤加味治疗后变化。方法:计算机采集分析系统记录和分析结肠电活动、运动和体表电活动信号;用免疫组织化学方法显示结肠神经细胞和神经纤维内 nNOS。结果:结肠肌层中 nNOS 阳性神经细胞和神经纤维增多,运动频率降低。加味四君子汤治疗后各指标基本恢复至正常水平。体表结肠电活动与慢波的峰值频率相关性比较中,正常组、治疗组相关性皆明显高于其他各组。结论:在脾虚状态下,结肠环行肌运动频率降低,容易造成溏便;nNOS 增加对运动频率降低有影响,但机体仍存在调节稳态的机制。加味四君子汤对脾虚证结肠动力紊乱有一定调理作用。

Abstract:Objective:To investigate the change of colinic circular muscle motility,electrical activity,cutaneous electrical activity and neuronal nitric oxide synthase(nNOS) in Spleen Deficiency syndrome (SDS) rats and the restoration after treatment.Methods:The motility and electrical signals were recorded and analyzed by computer.The colon nerve cells and nNOS is displayed by immunohistochemistry method.Results:In SD rats the frequency of colonic circular muscle motility decreased,nNOS positive nerve cells and nerve fibers in colonic circular muscle tissue increased.Most of the parameters returned to normal level after treated with modified Sijunzi Decoction.In stressed rats,the frequency and amplitude of the motility elevated,the peak frequency correlation of slow wave and cutaneous electrical activity was high in normal and Sijunzi Decoction treated rats.Conclusion:In SDS rats,the reduced frequency of colonic circular muscle could induce diarrhea;however,the body still keep in homeostasis;Sijunzi Decoction was effective on the colon motility disturbances; SDS induced systemic complicated disorders including pathological change of muscle,nerve system and so on,which could affect cuta- neous electrical activity that lead to the correlation of slow wave and cutaneous electrical activity changes.
