山茱萸水溶物对老年小鼠淋巴细胞功能的影响

摘    要:目的:研究山茱萸水溶物对老年小鼠的免疫调节作用,探讨其延缓衰老的机理。方法:采用氚标记胸腺嘧啶脱氧核苷(~3H-TdR)掺入法测定淋巴细胞的增殖;以 K562细胞为靶细胞测定自然杀伤细胞(NK)细胞活性;~3H-缬氨酸掺入法测定总蛋白合成;以依赖株细胞增殖反应测定法检测白细胞介素-2(IL-2)活性;用32P-IL-2 cDNA 探针,通过胞浆印迹杂交法测定 IL-2 mRNA 含量。结果:老年小鼠中,山茱萸水溶物给药组与对照组比较,淋巴细胞的增殖指数增加18.2%(P<0.01);NK 活性提高30.6%(P<0.01);总蛋白合成率增加13.9%(P<0.01);IL-2活性提高14.9≤(P<0.05);并可影响 IL-2 mRNA 的表达。结论:山茱萸水溶物显著提高老年小鼠淋巴细胞增殖能力、NK 活性、总蛋白合成、IL-2活性以及 IL-2 mRNA 表达水平。山茱萸水溶物能够部分恢复老年小鼠的免疫功能。

Effect of Water Extract of Fructus Comus on Function of Lymphocyte in Aged Mice

Abstract:Objective:To study the immune regulation effect of water extract of Fructus Comus (FC) in aged mice to explore its mechanism in retarding aging.Methods:After ConA-stimulation,the proliferation response of lymphocytes,NK cell activity and total protein syn- thesis capability of lymphocy-tes were determined by ~3H-TdR incorporation,~3H-TdR incorporating K562 cell line and ~3H-Valine incor- poration respectively.CTLL-20 cell (IL-2 dependent cell line) proliferation assay was used to determine IL-2 activity in ConA-stimulat- ed lymphocytes.IL-2 mRNA expression was measured by dot blotting.Results:After FC treatment,the stimulation index,NK activity, total protein synthesis and IL-2 activity were increased by 18.2%,30.6%,13.9% and 14.9% respectively,as compared with the ag- ing control.The dot blotting result showed that the expression level of IL-2 mRNA was also enhanced in FC treated mice.Conclusion: FC markedly enhanced lymphocyte proliferation,NK activity,total protein synthesis,IL-2 activity and IL-2 mRNA expression level, suggesting that FC has partly restorative effects on immune system associated with aging.
