芍药苷提取工艺和抗氧化活性

摘    要:目的 探索赤芍抗氧化活性组分最佳提取工艺,并对其有效组分芍药苷进行含量测定。方法 采用超声波提取法对赤芍中抗氧化活性组分进行提取,筛选最佳提取方案;利用硅胶柱层析对有效组分进行分离纯化,高效液相色谱法测定芍药苷含量。结果 选用浓度为70%的乙醇溶液,按照1∶30的料液比对赤芍进行超声50 min的提取,正丁醇萃取后通过硅胶柱层析可得到浓度为2.34%的芍药苷;正丁醇萃取得到产物的抗氧化活性最好。结论 超声提取后分离纯化的芍药苷具有较强的抗氧化活性。

Abstract:Objective To determine the contents of Paeoniflorin in radix paeoniae rubra, and to analyze the antioxidant activities in vivo.Methods Paeoniflorin, an antioxidant component in radix Paeoniae rubra, was extracted by ultrasonic extraction method. An extraction agent with strong antioxidant activity was selected through in vitro antioxidant experiments. The effective component was further separated and purified by silica gel column chromatography. Paeoniflorin content was determined by HPLC.Results Seventy percent ethanol solution was selected and ultrasonic extracting was conducted from radix paeoniae rubra for 50 minutes according to the ratio of material to liquid of 1∶30. After extracting with n-butanol, silica gel column chromatography was used to obtain 2.34 % paeoniflorin. The in vitro experiments of the extracted part proved that the antioxidant activity of the product obtained by n-butanol extraction was the most optimal.Conclusion Paeoniflorin isolated and purified after ultrasonic extraction has been proved to have antioxidant effects in vitro experiments.
