黄芪糖蛋白通过调控Nrf2/NQO1信号通路保护急性脊髓损伤大鼠模型神经功能

摘    要:目的 探讨黄芪糖蛋白对急性脊髓损伤大鼠模型的神经保护功能及机制。方法 选取30只SD大鼠,随机分为3组:假手术组、模型组及观察组。观察黄芪糖蛋白对大鼠肢体运动功能的影响,检测各组大鼠脊髓组织肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、髓过氧化物酶(myeloperoxidase,MPO)、白细胞介素-6(interleukin-6,IL-6)、核因子E2相关因子2(nuclear factor erythroid-2 related factor 2,Nrf2)、醌氧化还原酶1(quinone oxidoreductase,NQO1)表达水平。结果 观察组、模型组大鼠运动功能评分低于假手术组(P<0.05);观察组评分高于模型组(P<0.05)。观察组、模型组大鼠TNF-α、MPO、IL-6水平高于假手术组(P<0.05);观察组炎性因子水平低于模型组(P<0.05)。观察组、模型组大鼠脊髓组织Nrf2、NQO1mRNA表达水平低于假手术组(P<0.05);观察组表达水平高于模型组(P<0.05)。模型组与观察组Nrf2、NQO1蛋白量降低;观察组高于模型组(P<0.05)。结论 黄芪糖蛋白可通过上调脊髓组织Nrf2、NQO1表达水平发挥急性脊髓损伤大鼠神经保护功能。

Abstract:Objective To investigate the neuroprotective function and underlying mechanism of Huangqi glycoprotein in rats with acute spinal cord injury.Methods Thirty SD rats were randomly assigned into 3 groups,sham operation group,model group and observation group with 10 rats in each group.Then the motor function,and the expression levels of tumor necrosis factor-α (TNF-α),myeloperoxidase (MPO),interleukin-6 (IL-6),nuclear factor erythroid-2 related factor 2 (Nrf2) and NAD(P)H quinone oxidoreductase 1 (NQO1) were compared among three groups.Results The motor function scores after 1,2 and 4 weeks of intervention among three groups were the lowest in model group,followed by observation group,and were the highest in sham operation group,with statistical difference (P< 0.05).The levels of TNF-α,MPO and IL-6 of observation group and model group were higher than those of sham operation group (P<0.05),and the levels of the three indicators of observation group were lower than those of model group (P<0.05).The mRNA expression levels of Nrf2 and NQO1 in spinal cord tissues among three groups were the highest in sham operation group,followed by observation group,and were the lowest in model group,with statistical difference (P<0.05).The protein expression levels of Nrf2 and NQO1 in the spinal cord tissues of observation group and model group were lower than those of sham operation group (P<0.05),and the protein expression levels of observation group were lower than those of model group (P<0.05).Conclusion Huangqi glycoprotein exerts neuroprotective function in rats with acute spinal cord injury by upregulating the expression levels of Nrf2 and NQO1 in spinal cord tissues.
